AMG-966 is a human bispecific antibody (bsAb) targeting TNFSF15/TL1A & TNFα. AMG 966 restores the ability of the aglycosylated Fc domain to bind FcγRIa and FcγRIIa, leading to the formation of anti-drug antibodies (ADA). AMG 966 can be used in research on Crohn's disease and ulcerative colitis. Recommended isotype control: Human IgG1 kappa, Isotype Control (HY-P99001).

AMG-966 (300 nM; 6 days) elicits minimal CD4+ T cell proliferation in a human DC:T cell assay from 30 global HLA-representative donors, with no impact from its engineered charge pair mutations[1].
AMG-966 (1:1, 1:1:1, 9:1:1, 200:1:1, 600:1:1 molar ratios; approximately 16 hours) forms intermediate-sized immune complexes with recombinant human TNF at a 1:1 ratio and larger complexes when combined with both TNF and TL1A at lower drug-to-target ratios, as measured by SEC-MALS after 16-hour incubation at 4°C[1].
AMG-966 (1:1:1, 4:1:1, 9:1:1, 100:1:1 molar ratios; overnight) forms immune complexes with recombinant human TNF and TL1A that exhibit enhanced binding to FcγRIa and FcγRIIa at low 1:1:1 drug-to-target ratios (after overnight incubation at 4°C), with binding decreasing as the drug-to-target ratio increases, while FcγRIIIA binding is minimally affected[1].

Flow cytometric analysis of 1×106 MCF-7 cells labelling TL1A (red) with AMG-966 (HY-P991440). Cells were fixed with 4% paraformaldehyde and permeabilised with 90% methanol. Then stained with the primary antibody at 1/200 dilution for an hour at 4℃. Alexa Goat Anti-Human IgG H&L (AF488) (HY-P83776) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Human IgG1 kappa Isotype Control (HY-P99001, blue) was used as the isotype control.

DSS-Induced Colitis
Dextran sulfate sodium (DSS)-induced colitis is generated by administering DSS in mouse drinking water, producing epithelial injury, barrier disruption, weight loss, diarrhea, fecal blood, colon shortening, histologic mucosal damage, and inflammatory mediator changes; the model is mainly used to study acute or chronic intestinal inflammation resembling selected features of ulcerative colitis. DSS injury is interpreted through clinical and tissue readouts rather than a single molecular endpoint: daily body weight, stool consistency, and bleeding are combined into a disease activity index, while colon length, histology, cytokines, myeloperoxidase activity, intestinal permeability, and tight-junction markers provide complementary measures of inflammation and barrier damage.

TNBS-Induced Colitis
TNBS-induced colitis is produced by intrarectal delivery of 2,4,6-trinitrobenzene sulfonic acid in ethanol, where ethanol disrupts the mucosal barrier and TNBS haptenates colonic proteins, generating immune-mediated colonic inflammation with weight loss, diarrhea, ulceration, transmural injury, inflammatory-cell infiltration, and cytokine responses. The model is used as an experimental intestinal inflammation model with Crohn’s disease–like features, especially when Th1-type responses, IL-12–dependent inflammation, chronic relapsing inflammation, or fibrosis-related endpoints are studied.
