Insulin lispro
Based on 1 Customer Validation
Insulin lispro is a recombinant h-Insulin (HY-P0035) analog whose primary structure is identical to regular h-Insulin, except that the proline-lysine at positions B28 and B29 is translocated to lysine-proline, thereby reducing self-association. Insulin lispro stimulates insulin receptor internalization to early endosomes, induces receptor phosphorylation, intracellular internalization, and colocalization with the early endosome marker EEA1. Insulin lispro shows stronger inhibition of hepatic glucose production and stronger inhibition of glucagon secretion. Insulin lispro has lower mitogenic potency. Insulin lispro can be used in the study of diabetes.

In Vitro
Insulin lispro (5 nM; 5-60 min) induces internalization of phosphorylated IRβ (Tyr1146) in U2OS-hIR cells and colocalizes with early endosomes at 5 to 60 minutes[1].
Insulin lispro (100-≥580 μU/mL) does not cross the placenta at concentrations up to 200 μU/mL, but a small concentration-dependent transfer occurs at concentrations ≥580 μU/mL[2].
MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.

Result:Produced a distinct punctate intracellular staining pattern for IRβ compared with control cells.
Showed overlap of EEA1 with the majority of the internalized IRβ signal.
Induced internalized IRβ starting at 5 minutes and throughout the time-course up to 60 minutes.
Showed no overlay of LAMP1 staining with IRβ staining before or after treatment.\nProduced a distinct punctate intracellular staining pattern for phospho-IRβ with no significant immunostaining at the cell surface.
Induced internalized phospho-IRβ starting at 5 minutes and throughout the time-course up to 60 minutes.
Showed overlap of EEA1 with most of the phospho-IRβ signal.
Showed a low level of co-localization of phospho-IRβ with lysosomes.
