Cangrelor (AR-C69931MX), an adenosine triphosphate analogue, is an intravenous, reversible and selective platelet P2Y12 antagonist, with prompt and potent antiplatelet effects. Cangrelor directly blocks adenosine diphosphate (ADP)-induced activation and aggregation of platelets. Cangrelor is also a nonspecific GPR17 antagonist.

The free form of the compound is prone to instability, it is advisable to consider the stable salt form (Cangrelor tetrasodium) that retains the same biological activity.

    The EA. hy926 cells were pretreated with vehicle (M), OG-5 for 2 hours followed by stimulated with ADP for 4 hours, then the treated cells and supernatants were collected. The release of MCP-1 and VCAM-1 in supernatant were determined by ELISA kit and adhesion molecule ICAM-1 was determined in cell lysates. The results showed that treatment with OG-5 significantly inhibited the increase in ADP-induced MCP-1 and VCAM-1 production, showing a comparable effect with Cangrelor tetrasodium (0.5 µM) at the concentration of 0.5 mM.

    Immunoblotting and densitometric analyses of p-VASP (Ser157). Platelets were preincubated with vehicle and different concentrations of OG-5 for 5 min at 37 ℃, in the presence or absence of the specific inhibitors and then stimulated with agonist. The reaction was stopped by addition of RIPA buffer immediately and the cell lysates were applied to immunoblot analysis with phospho-specific antibody. The results showed that Cangrelor tet (0.5 μM) attenuated the 2MeS-ADP-induced reduction in p-VASP(S157) levels.

    In Vivo

    Cangrelor tetrasodium (10 mg/kg) not only significantly decreases BLM-induced release of inflammatory cytokines (PF4, CD40 L and MPO), but also decreases the increment of platelets, neutrophils and platelet-neutrophil aggregates in the fibrotic lung and in the peripheral blood of BLM-treated mice[2].
