Neorauflavane

Neorauflavane is a tyrosinase inhibitor with mushroom tyrosinase IC50 values of 30 nM and 500 nM, and Ki values of 19 nM and 130 nM. Neorauflavane inhibits monophenolase and diphenolase activities, binds to tyrosinase’s active site via its B-ring resorcinol motif and A-ring methoxy group. Neorauflavane reduces melanin synthesis in melanoma cells. Neorauflavane can be used for the research of skin hyperpigmentation.

Neorauflavane (Compound 3) (0-140 nM; 10-25 min) potently inhibits the monophenolase activity of mushroom tyrosinase via a competitive, simple reversible slow-binding inhibition mode, with an IC50 of 30 nM and a Ki of 19 nM[1].
Neorauflavane (0-720 μM; 25 min) inhibits the diphenolase activity of mushroom tyrosinase via competitive inhibition, with an IC50 of 500 nM and a Ki of 130 nM[1].
Neorauflavane (0-20 μM; 72 h) reduces α-MSH (HY-P0252)-stimulated melanin content in B16 mouse melanoma cells in a dose-dependent manner, with an IC50 of 13 μM, and shows no cytotoxicity at effective concentrations[1].
