Cedryl acetate is an orally active α-glucosidase inhibitor with an IC50 of 94 μM against yeast α-glucosidase. Cedryl acetate reduces high-fat diet-induced body weight gain, visceral fat pad weight, adipocyte hypertrophy, hepatic lipid accumulation, glucose intolerance, insulin resistance and gluconeogenesis. Cedryl acetate can be used in the research of obesity and obesity-related metabolic syndrome.

Animal Model:C57BL/6J (male, 7 weeks old at study start, acclimatized for 3 weeks prior to treatment, obesity induced by 40% fat high-fat diet)[1]

Result:Reduced final body weight to 33.68 g (vs. 46.61 g in HFD-only mice) and body weight gain to 8.93 g (vs. 21.69 g in HFD-only mice).
Significantly decreased total visceral fat pad weight, and reduced mean epididymal adipocyte cross-sectional area to 2035 µm2 (vs. 4275 µm2 in HFD-only mice).
Lowered serum levels of LDL-C to 0.72 mmol/L (vs. 1.49 mmol/L in HFD-only mice), HDL-C to 4.83 mmol/L (vs. 6.57 mmol/L in HFD-only mice), total cholesterol to 4.70 mmol/L (vs. 8.57 mmol/L in HFD-only mice), and ALT to 61.4 U/L (vs. 237.3 U/L in HFD-only mice).
Improved glucose homeostasis: reduced fasting blood glucose, lowered blood glucose levels at 60, 90, and 120 minutes during oral glucose tolerance testing (with reduced AUC), and lowered blood glucose levels at 15, 30, 60, and 90 minutes during insulin tolerance testing (with reduced AUC).
Inhibited hepatic gluconeogenesis: reduced blood glucose levels at 30, 60, 90, and 120 minutes during pyruvate tolerance testing (with reduced AUC), and downregulated hepatic mRNA expression of gluconeogenesis-related genes Pepck and G6Pase.
Reduced liver weight and attenuated hepatic lipid accumulation.
Modulated eWAT gene expression: downregulated mRNA levels of adipogenesis/lipid synthesis genes PPARγ, C/EBPα, FABP4, and FAS; upregulated mRNA levels of thermogenesis-related genes PGC-1α, Cidea, Cytc, COX4, and PRDM16.
Showed no significant differences in gut microbiota α-diversity indexes, β-diversity clustering, or phylum/family-level composition compared to HFD-only mice.

- To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for In Vivo experiments, it is recommended to prepare freshly and use it on the same day.
- The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.
Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline

Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
Add each solvent one by one: 10% DMSO 90% (20% SBE-β-CD in Saline)
Solubility: ≥ 2.5 mg/mL (9.46 mM); Clear solution
This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.
Preparation of 20% SBE-β-CD in Saline (4°C, storage for one week): 2 g SBE-β-CD powder is dissolved in 10 mL Saline, completely dissolve until clear.
