N-Acetyltryptamine (N10-Acetyltryptamine) is a mixed agonist-antagonist of the melatonin receptor. N-Acetyltryptamine inhibits acetylserotonin methyltransferase, blocks the conversion of N-acetylserotonin to melatonin, and inhibits neuronal firing activity in suprachiasmatic nucleus neurons. N-Acetyltryptamine is a physiological constituent of mammalian blood. N-Acetyltryptamine is a metabolite of tryptamine in Bacillus cereus.

N-acetyltryptamine is present in daytime plasma of rats, rhesus macaques, and humans, establishing it as a physiological constituent of mammalian blood[1].
N-acetyltryptamine (100 μM-10 mM) exhibits concentration-dependent mixed agonist-antagonist effects on Melatonin (HY-B0075)-evoked responses in rat SCN neurones, acting as an antagonist at 100 μM (blocking Melatonin-evoked responses with a smaller reduction in firing) and as an agonist at 1-10 mM (suppressing neuronal discharge)[2].
N-Acetyltryptamine acts as a noncompetitive inhibitor versus Tryptamine (HY-B2132) and an uncompetitive inhibitor versus Acetyl-CoA (Acetyl coenzyme A) (HY-114293) in the GST-AANAT-catalyzed reaction, supporting an ordered BiBi ternary complex mechanism where N-acetyltryptamine is the first product released[3].
N-Acetyltryptamine is formed as an artifact during the extraction of Tryptamine with ethyl acetate under mild basic conditions via base-catalyzed aminolysis, as confirmed by TLC, mass spectrometry, and comparison with the authentic synthetic complex[4].
N-Acetyltryptamine during senescence completely inhibits melatonin synthesis without altering N-acetylserotonin levels in detached Asahi rice leaves[5].
N-Acetyltryptamine is a genuine metabolite of tryptamine in Bacillus cereus, not an extraction artifact[4].

Select the appropriate dissolution method based on your experimental animal and administration route.
- For the following dissolution methods, please ensure to first prepare a clear stock solution using an In Vitro approach and then sequentially add co-solvents:
- To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for In Vivo experiments, it is recommended to prepare freshly and use it on the same day.
- The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.
Add each solvent one by one: 10% EtOH 40% PEG300 5% Tween-80 45% Saline
Solubility: ≥ 5 mg/mL (24.72 mM); Clear solution
This protocol yields a clear solution of ≥ 5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL EtOH stock solution (50.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
Add each solvent one by one: 10% EtOH 90% (20% SBE-β-CD in Saline)
Solubility: ≥ 5 mg/mL (24.72 mM); Clear solution
This protocol yields a clear solution of ≥ 5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL EtOH stock solution (50.0 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.
Preparation of 20% SBE-β-CD in Saline (4°C, storage for one week): 2 g SBE-β-CD powder is dissolved in 10 mL Saline, completely dissolve until clear.
Add each solvent one by one: 10% EtOH 90% Corn Oil
Solubility: ≥ 5 mg/mL (24.72 mM); Clear solution
This protocol yields a clear solution of ≥ 5 mg/mL (saturation unknown). If the continuous dosing period exceeds half a month, please choose this protocol carefully.
Taking 1 mL working solution as an example, add 100 μL EtOH stock solution (50.0 mg/mL) to 900 μL Corn oil, and mix evenly.
