Description

cTMTP1 (Cyclic TMTP1) is the cyclic form of TMTP1 (HY-P11815). TMTP1 is a tumor-targeting peptide that selectively targets highly metastatic tumor cells. The cTMTP1-based [68Ga]Ga-DOTA-cTMTP1 probe is used for visualization of hepatocellular carcinoma. cTMTP1 can form a 68Ga-DOTA radiotracer that accumulates in highly metastatic cervical cancer cells and is used for microPET tumor visualization. cTMTP1 is applicable to research related to hepatocellular carcinoma and cervical cancer[1][2].

In Vitro

cTMTP1-based radiotracer 68Ga-DOTA-TMTP1 (~18.5 KBq/well; 15-120 min; 10 μg cold cyclic TMTP1 pre-incubated for 15 min followed by 60-120 min incubation) exhibits specific, high uptake and retention in highly metastatic HeLa cervical cancer cells compared to low metastatic C-33A cells and normal End1 cells[2].

In Vivo

[68Ga]Ga-DOTA-cTMTP1 (5.2 MBq; i.v.; single dose) exhibits sustained tumor retention and high tumor-to-liver ratios (ranging from 2.14 to 2.69) in subcutaneous HCC xenograft mice, demonstrating specific targeting and favorable pharmacokinetics for HCC imaging[1].
[68Ga]Ga-DOTA-cTMTP1 (i.v.; single dose) maintains high tumor-to-liver ratios (ranging from 2.20 to 2.55) and enables sensitive detection of small HCC lesions in transgenic c-Myc liver tumor mice[1].

Animal Model: BALB/c nude (male, 5−6 weeks old, ~20 g, subcutaneous xenograft via right forelimb axillary injection of 2×106 SMMC-7721 cells)[1]
