Polzastobart (JTX-8064) is a humanized IgG4 monoclonal antibody and LILRB2 (ILT4) antagonist. Polzastobart specifically binds to and antagonizes LILRB2, relieves ILT4-mediated myeloid immunosuppression, and promotes the polarization of macrophages from an M2-like phenotype to an M1-like phenotype. Polzastobart induces macrophage polarization, IFNγ signaling, and T cell inflammation-related pharmacodynamic responses in ex vivo human renal cancer, lung cancer, and head and neck cancer tissues. Polzastobart can be used in studies related to tumor-associated macrophages and tumor immunity. Polzastobart (JTX 8064) repolarizes macrophages from the immunosuppressive M2 phenotype to the pro-inflammatory M1 phenotype, thereby enhancing anti-tumor activity[3].
Polzastobart (administered for 24 h) induces a pharmacological response superior to that of the isotype control in more than 100 fresh, untreated ex vivo tissue culture samples of human renal cell carcinoma, lung cancer, and head and neck cancer, characterized by macrophage polarization, IFNγ signaling, and T cell inflammation-related features[2].

Flow cytometric analysis of 1X106 THP-1 cells with Polzastobart (HY-P990061, red). Cells were fixed with 4% paraformaldehyde. Then stained with the primary antibody at 1/200 dilution for an hour at 4℃. Alexa Fluor 488-conjugated AffiniPure Goat Anti-Human IgG H&L (HY-P83776) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Human IgG4 kappa (HY-P99003, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black). -
Immobilized LILRB2/CD85d/ILT-4 Protein, Human can bind Polzastobart. The EC50 for this effect is 8.336 ng/mL.
