Polzastobart (JTX-8064) is a humanized IgG4 monoclonal antibody and LILRB2 (ILT4) antagonist. Polzastobart specifically binds to and antagonizes LILRB2, relieves ILT4-mediated myeloid immunosuppression, and promotes the polarization of macrophages from an M2-like phenotype to an M1-like phenotype. Polzastobart induces macrophage polarization, IFNγ signaling, and T cell inflammation-related pharmacodynamic responses in ex vivo human renal cancer, lung cancer, and head and neck cancer tissues. Polzastobart can be used in studies related to tumor-associated macrophages and tumor immunity.     Flow cytometric analysis of 1X106 THP-1 cells with Polzastobart (HY-P990061, red). Cells were fixed with 4% paraformaldehyde. Then stained with the primary antibody at 1/200 dilution for an hour at 4℃. Alexa Fluor 488-conjugated AffiniPure Goat Anti-Human IgG H&L (HY-P83776) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Human IgG4 kappa (HY-P99003, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black).
