XMU-MP-9 is a bifunctional compound that binds to the C2 domain of Nedd4-1 and the allosteric site of K-Ras. XMU-MP-9 enhances the interaction between Nedd4-1 and K-Ras, induces conformational changes in the Nedd4-1/K-Ras complex, promotes the ubiquitination and degradation of multiple K-Ras mutants, and inhibits the proliferation of cells carrying K-Ras mutants. XMU-MP-9 can be used for the study of colon, lung and pancreatic cancer.

Biologische Aktivität
XMU-MP-9 (10 μM; 48 h) promotes the degradation of K-RasG12V via the lysosomal pathway in SW620 cells[1].
XMU-MP-9 (2-20 μM) inhibits the proliferation of cancer cells (SW620, AsPC-1, etc.) carrying K-Ras mutants in 2-D culture[1].

Animal Model:BALB/c mice with CT-26 transplant tumors[1]

Dosage:40 mg/kg, 80 mg/kg

Administration:Tail vein injection; once or twice daily

Result:Showed more robust inhibitory effect on CT-26 transplant tumors in BALB/c mice.
Decreased K-RasG12V levels and phosphorylation of B-Raf and MEK in tumors.
Showed no significant weight loss or toxicity to heart, liver, spleen, lung, or kidney.
