Cibisatamab, a T cell bispecific antibody, binds Carcino-Embryonic Antigen (CEA) on cancer cells and CD3 on T cells. Cibisatamab triggers T cell killing of cancer cell lines expressing moderate to high levels of CEA at the cell surface. Cibisatamab can be used for colorectal cancer research.

Cibisatamab (20 nM; co-cultured with CD8 T cells for 7-10 days) is highly sensitive to patient-derived colorectal cancer organoids (PDOs) with high CEA expression, shows resistance to PDOs with low CEA expression, and has low sensitivity to PDOs with mixed CEA expression[1].

Flow cytometric analysis of 1X106 Jurkat cells with Cibisatamab (HY-P99011, red). Cells were fixed with 4% paraformaldehyde. Then stained with the primary antibody at 1/200 dilution for an hour at 4℃. Alexa Fluor 488-conjugated AffiniPure Goat Anti-Human IgG H&L (HY-P83776) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Human IgG1 kappa (HY-P99001, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black).
