Description

Elotuzumab (HuLuc 63) is an IgG1 monoclonal antibody targeting the SLAMF7 receptor. Elotuzumab exerts antitumor activity by activating natural killer cells and inducing antibody-dependent cell-mediated cytotoxicity. Elotuzumab can be combined with Lenalidomide (HY-A0003), Dexamethasone (HY-14648), etc., for the research of tumors such as multiple myeloma[1][2].

In Vitro

Elotuzumab (0.1-10 μg/mL; 4-20 h) upregulates the expression of CD107a, CD69 and CD54 on NK cells, and promotes the secretion of IFN-γ and TNF-α in a co-culture system of human PBMC and OPM-2 cells[2].

In Vivo

Elotuzumab (10 mg/kg; intraperitoneal injection; 3 times; once every 3-4 days) exhibits antitumor activity in a mouse tumor model expressing hSLAMF7, and the effect is better when used in combination with anti-PD-1 antibody[2].

Animal Model: Female BALB/c mice treated A20-hSLAMF7 cells or EG7-hSLAMF7 cells[2]
Dosage: 10 mg/kg (Elotuzumab in the form of murine IgG2a subtype)
Administration: Intraperitoneal injection; 3 times; once every 3-4 days
Result: Inhibited tumor growth, increased tumor-infiltrating NK cells and CD8+ T cells, and the expression of activation markers on these cells.
Promoted tumor-infiltrating NK and CD81 T-cell activation, as well as increased intratumoral cytokine and chemokine release by combination with anti-PD-1 antibody.
