Eschweilenol C (72 h) potently inhibits the growth of PfDd2, Pf3D7, and mutant-resistant PfDd2-GNF156 Plasmodium falciparum asexual blood stages with IC50 values ranging from 379.21 nM to 490.74 nM and a low resistance index of 1.29[2].
Eschweilenol C (1-5 μg/mL; 48 h) is inactive against late-stage IV/V Plasmodium falciparum NF54-E gametocytes, with an IC50 greater than 5 μg/mL[2].
Eschweilenol C is non-toxic to Vero, Raw 264.7, and O+ human red blood cells, with high selectivity indices (SI > 10) against Plasmodium falciparum asexual blood stages[2].
Eschweilenol C (72 h) significantly inhibits haemozoin production by Plasmodium falciparum at its PfDd2 IC50 concentration of 490.74 nM[2].
Eschweilenol C (24-72 h) acts slowly against Plasmodium falciparum PfDd2, with an IC50 of 557.66 nM at 48 h and no measurable IC50 at 24 h[2].
Eschweilenol C preferentially inhibits Plasmodium falciparum PfDd2 ring and trophozoite stages (82.13% and 85.01% growth inhibition, respectively) with a parasiticidal effect, and has weaker activity against schizonts (58.88% growth inhibition) with a parasitostatic effect[2].
Eschweilenol C displays strong binding affinity with Plasmodium falciparum PfM1AAP (-8.7 kcal/mol) and PfM17LAP (-8.3 kcal/mol)[2].
