FSBA (5'-p-Fluorosulfonylbenzoyladenosine) hydrochloride is a covalent modifier and affinity labeling reagent for adenine nucleotide-binding proteins. FSBA hydrochloride covalently attaches to the nucleotide-binding sites of pyruvate kinase, glutamate dehydrogenase, and p56lck, and to a lysine residue in the ATP-binding site of cAMP-dependent protein kinase, causing loss of enzymatic activity. FSBA hydrochloride can be used for the research of T lymphoma.

Biological Activity
FSBA (2.5 mM; 2 h) hydrochloride covalently modifies bovine liver glutamate dehydrogenase at a stoichiometry of 1.4 mol FSBA per mol enzyme, and the modified protein is specifically detected by affinity-purified anti-FSBA antibodies on immunoblots[1].
FSBA (2.5 mM; 2 h) hydrochloride covalently modifies rabbit muscle pyruvate kinase, and the modified protein is specifically detected by affinity-purified anti-FSBA antibodies on immunoblots[1].
FSBA (0.9 mM; 0-40 min) hydrochloride covalently modifies the bovine heart catalytic subunit of cAMP-dependent protein kinase in a time-dependent manner, resulting in proportional loss of kinase activity, with modified enzyme specifically detected by affinity-purified anti-FSBA antibodies[1].
FSBA (0.9 mM; 30 min) hydrochloride covalently modifies multiple nucleotide-binding proteins in cytosolic fractions from murine splenocytes, with labeling blocked by competing MgATP, and only minimally affected by increasing Mn2+ concentrations[1].
FSBA (0.125 mM; 25 min) hydrochloride covalently modifies the p56lck protein-tyrosine kinase in postnuclear particulate fractions from LSTRA cells, with far greater labeling observed in p56lck-overexpressing LSTRA cells than in SL-2 cells, and labeling blocked by competing MgATP[1].
FSBA (10-400 μM; 20 min) hydrochloride binds in a saturable manner to the p56lck protein-tyrosine kinase in postnuclear particulate fractions from LSTRA cells, confirming specific interaction with its nucleotide-binding site[1].
