CP61

CP61 is a CtBP1 and CtBP2 inhibitor that binds to dimeric CtBP1 and allosterically inhibits NADH (HY-113355) binding, disrupting the NADH-dependent dimerization of CtBP1 and CtBP2, inducing aberrant mitosis and micronucleus formation, thereby inhibiting cell proliferation and colony formation. CP61 can be used in breast cancer research.

CP61 introduces a cysteine by mutating its own SGW sequence to CGW and is conjugated via a disulfide bond to the N-terminally Cys-modified HIV TAT cell-penetrating peptide (GRKKRRQRRRPPQ), thereby achieving intracellular delivery of the cyclic peptide. The resulting CP61-TAT retains CFP-CtBP1 in the cytoplasm of COS-7 cells without altering total CtBP1/CtBP2 protein levels; it dose-dependently increases the proportion of micronuclei in MCF-7 cells, raises the fraction of abnormal mitosis and prolongs mitotic duration, and inhibits MCF-7 cell proliferation, and this effect is reproducible in ER-negative highly metastatic MDA-MB-231 cells^[1].

CP61 (50 μM; microinjection) selectively increases abnormal mitosis and prolongs mitotic duration in highly glycolytic MCF-7 cells, but has no such effect in MDA-MB-453 cells with lower glycolysis^[1].

Others

CP61 inhibits CtBP2 homodimerization in ELISA assays, IC₅₀ = 19 μM^[1].
CP61 dose-dependently disrupts the GST-CtBP1/His-CtBP1 complex in the presence of 10 μM NADH^[1].
CP61 allosterically and dose-dependently reduces the CtBP1-NADH FRET signal (Trp 285 nm, NADH 425 nm), and the maximal FRET is suppressed under saturating NADH, suggesting that it does not occupy the NADH pocket^[1].
CP61 shows no inhibitory effect on other Rossmann fold-containing NAD⁺/NADH-dependent dehydrogenases at concentrations up to 100 μM^[1].
