Adhumulone
Adhumulone is an α-acid derived from hops and a selective AKR1B10 inhibitor, with IC50 values of 5.41-29.27 μM and Ki values of 3.27-16.79 μM against human AKR1B10. Adhumulone inhibits AKR1B10-mediated reduction of Farnesol (HY-Y0248A) as a competitive inhibitor, and it also suppresses the reduction of DL-Glyceraldehyde (HY-128748) and Farnesal substrates. Adhumulone is useful for research on ras-dependent malignancies.

In Vitro
Adhumulone (Compound 1) selectively inhibits recombinant human AKR1B10 when DL-Glyceraldehyde (HY-128748) is used as the substrate, with an IC50 of 5.41 µM and a Ki value of 3.27 μM, while its activity against AKR1A1 and AKR1B1 is much weaker[1].
Adhumulone acts as a competitive inhibitor of recombinant human AKR1B10-mediated reduction of Farnesol (HY-Y0248A), with an IC50 of 29.27 µM and a Ki of 16.79 µM[1].
Adhumulone (41c) is a major α-acid sensory metabolite in fresh Humulus lupulus L. hop pellets, with concentrations ranging from 5-15% across 75 commercial hop varieties[2].
