In Vitro

CD27 peptide (99:1 peptide:siRNA ratio; 1 h) incorporation into HIF2α-CD27 PAMs successfully retains HIF2α siRNA and protects it from RNase degradation^[1].

In Vivo

Animal Model:

C57BL/6 (Female human CD27 transgenic (hCD27), aged 6-12 weeks)^[2]
Dosage:

100 µg
Administration:

i.p.; on days 3 and 6 after tumor implantation
Result:

Increased Ova(I)-specific CD8+ T cells from ~20% to ~50% of the CD8+ TIL population.
Extended median survival by 18 days compared to control animals.\nProlonged survival in the setting of vaccines comprised of linked class I and II epitopes (Ova(I-IIA)).
Had no effect on survival with Ova(I) or Ova(I) + Ova(IIA) vaccines.\nProlonged survival in hCD27 mice bearing intracranial B16.OVA tumors.
