NADs-IN-2 is a NADSYN1 inhibitor. NADs-IN-2 also binds to the NaAD subsite within the active site of bacterial NADs, thereby inhibiting enzymatic function. NADs-IN-2 can be used in research related to anthrax and cancer.

In Vitro
NADs-IN-2 (5824) (0.6-600 μM; 10 min) potently inhibits purified Bacillus anthracis NAD synthetase, with an IC50 value of 10 μM[1].
NADs-IN-2 (1.88-240 μM; 5 h) completely inhibits the vegetative growth of *Bacillus anthracis* Sterne strain, with an MIC100 of 1.9 μM[1].
NADs-IN-2 (NADSYNli) (72 h) selectively kills cancer cells with amplified PH pathway, while exerts no significant effects on normal cells and cancer cells dependent on the salvage pathway[2].
NADs-IN-2 (2 μM; 72 h) fails to induce drug sensitivity in non-cancer cells (IMR90, RPE-1) with NAPRT- or NAMPT-overexpression[2].
NADs-IN-2 (1-3 μM; 30 min) inhibits the activity of purified human NADSYN1 enzyme in a dose-dependent manner[2].

Result:Reduced the volume and intratumoral NAD+ levels of Preiss Handler pathway-dependent OV4PH-amp tumors in nude mice.
