PhoFAP-02

PhoFAP-02 is a photodynamic therapy photosensitizer targeting FAP, assembled from a FAPI-04-derived targeting moiety, the DOTA chelator, and the photosensitizer IRDye700DX (HY-D2309). PhoFAP-02 produces photodynamic cytotoxicity against FAP-positive cells under 690 nm light irradiation. PhoFAP-02 can be radiolabeled with indium-111 and accumulates in FAP-high cancer-associated fibroblast stromal regions in mouse models of pancreatic cancer. PhoFAP-02 can be used in research related to targeted photodynamic therapy of pancreatic ductal adenocarcinoma.

PhoFAP-02 binds to 3T3-FAP cells with FAP-mediated specificity^[1].
PhoFAP-02 (337.5-0.00075 nM; 2 h) inhibits the binding of [¹¹¹In]In-FAPI-04 to 3T3-FAP cells with an IC₅₀ of 27.75 nM^[1].
PhoFAP-02 (0-1 nM; 2 h) induces concentration-dependent phototoxicity in 3T3-FAP cells after 690 nm irradiation, while not damaging 3T3-WT cells and showing no dark toxicity^[1].
