Plantagoside is a flavanone glucoside found in Plantago asiatica seeds, acting as a specific non-competitive α-mannosidase inhibitor with IC50 values of 5 μM and a Ki of 2.7 μM (jack bean). Plantagoside suppresses antibody response and Concanavalin A (HY-P2149)-induced lymphocyte proliferation in mouse spleen cells. Plantagoside inhibits the Maillard reaction, advanced glycation end product formation, and glycation-dependent protein-protein cross-link formation. Plantagoside can be used for the research of diabetes.

Actividad biológica
Plantagoside potently and specifically inhibits jack bean α-mannosidase (IC50 = 5 μM), while showing negligible to weak inhibition of other tested glycosidases[1].
Plantagoside inhibits mouse liver lysosomal α-mannosidase (IC50 = 25 μM) and microsomal α-mannosidase (IC50 = 100 μM)[1].
Plantagoside (3-100 μg/mL; 4 days) dose-dependently suppresses anti-SRBC antibody formation in cultured mouse spleen cells[1].
Plantagoside (1-100 μg/mL; 2 days) dose-dependently suppresses Concanavalin A (HY-P2149)-induced proliferation of BALB/c mouse spleen cells, with no effect on PHA- or LPS (HY-D1056)-induced proliferation[1].
Plantagoside (30 h) potently inhibits Maillard reaction-mediated AGE formation in BSA at 60°C with an IC50 of 1.2 μM[2].
Plantagoside inhibits Maillard reaction-mediated AGE formation in collagen at 37°C with an IC50 of 14 μM[2].
Plantagoside (12.5-200 μM; 1 week) inhibits ribose-mediated dimerization of lysozyme in a concentration-dependent manner[2].

- To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for In Vivo experiments, it is recommended to prepare freshly and use it on the same day.
- The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.
Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline
Solubility: ≥ 2.5 mg/mL (5.36 mM); Clear solution
This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
Add each solvent one by one: 10% DMSO 90% (20% SBE-β-CD in Saline)
Solubility: ≥ 2.5 mg/mL (5.36 mM); Clear solution
This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.
Preparation of 20% SBE-β-CD in Saline (4°C, storage for one week): 2 g SBE-β-CD powder is dissolved in 10 mL Saline, completely dissolve until clear.
