SERTlight is a fluorescent agent that can specifically label serotonergic neuronal cell bodies, dendrites, and axonal projections as fluorescent substrates of the serotonin transporter (SERT). SERTlight is orthogonal to various genetically encoded sensors in terms of optics, pharmacology, and operation, enabling multiple imaging. SERTlight allows the use of GRAB5HT sensors to label distal 5HT axonal projections and simultaneously image the release of endogenous 5HT, providing a new multifunctional molecular tool for studying serotonergic systems .

HMRG is a fluorescent dye, which serves as the fluorescent active product generated by enzyme-activated fluorescent probes targeting proteases and glycosidases. Its detection mechanism relies on the intramolecular spirocyclization equilibrium: HMRG itself exists in an open non-spirocyclic structure under physiological pH, which maintains a conjugated π system and produces strong fluorescence; whereas its probe precursors (such as Leu-HMRG, Ac-GlyPro-HMRG and βGal-HMRG) exist in a closed spirocyclic form and show no fluorescence due to the disruption of the π-conjugated system; when the target enzyme cleaves the substrate fragment linked to the probe precursor, the closed spirocyclic structure converts to the open structure of HMRG, restoring the conjugated system and generating a strong fluorescent signal. Its detection wavelength is Ex/Em = 501/524 nm .

DABCYL-LPETG-EDANS is a biological active peptide. (This 5-amino acid peptide is a sortase substrate, C-terminal sorting signal. Sortase cleaves surface proteins at the LPXTG motif and catalyzes the formation of an amide bond between the carboxyl group of threonine and the amino group of cell-wall crossbridges. Sortases are a family of Gram-positive transpeptidases responsible for anchoring surface protein virulence factors to the peptidoglycan cell wall layer. Cleavage of this FRET substrate by sortase reveals the fluorescent signal, Abs/Em = 340/490 nm.)
