ZK53 is a selective activator of mitochondrial caseinolytic protease P (HsClpP) (EC50: 1.37?μM for α-casein hydrolysis by HsClpP). ZK53 is is inactive toward bacterial ClpP proteins. ZK53 induces apoptosis in H1703, H520 and SK-MES-1 cells. ZK53 induces dysregulation of mitochondrial functions in lung squamous cell carcinoma (LUSC) cells. ZK53 inhibits tumor growth in H1703 xenograft mouse model .

Cisd2 agonist 2 (compound 6) is a Cisd2 activator (EC50=191 nM), and Cisd2 levels are associated with non-alcoholic fatty liver disease (NAFLD). Cisd2 agonist 2 has no significant in vivo toxicity in Cisd2hKO-het mice (heterozygous hepatocyte-specific Cisd2 knockout). Cisd2 (CDGSH iron sulfur domain 2) is a zinc finger protein that is mainly localized in the endoplasmic reticulum or mitochondrial membrane. Cisd2 participates in mitochondrial function by forming homodimers containing two redox-active 2Fe-2S clusters .

Lisaftoclax is an orally active, selective BCL-2 inhibitor with a Ki of < 0.1 nM against human BCL-2. Lisaftoclax selectively binds to BCL-2, disrupts the BCL-2:BIM complex, and antagonizes the antiapoptotic function of BCL-2. Lisaftoclax induces BAX/BAK-dependent, caspase-mediated apoptosis, upregulates the pro-death proteins BIM and Noxa, and downregulates mitochondrial respiratory function and ATP production. Lisaftoclax can be used in the research of hematological malignancies including chronic lymphocytic leukemia, multiple myeloma, and diffuse large B-cell lymphoma .

4A7C-301 is a blood-brain barrier-permeable Nurr1 agonist, with IC50 values of 48.22 nM and 107.71 nM for binding to Nurr1-LBD in the [ 3H]-CQ competition assay and TR-FRET assay, respectively.
