Spesolimab (LALA)
Spesolimab (MAB92; BI 655130) (LALA) is a humanized IgG1k antibody against IL-36R with a Kd value of 20 pM. Spesolimab (LALA) selectively blocks IL-36R signaling without inhibiting related interleukin pathways. Spesolimab (LALA) lacks functional cross-reactivity with non-human IL-36R and exhibits species-specific binding to human IL-36R, with reduced affinity for mouse, rat, hamster, minipig, and non-human primate IL-36R. Spesolimab (LALA) has L234A and L235A mutations introduced into the Fc region, designed to reduce antibody binding to FcγR and C1q, thereby attenuating/eliminating the ADCC and CDC effector functions of the antibody. Spesolimab (LALA) can be used for research on generalized pustular psoriasis (GPP).

In Vitro
Spesolimab (LALA) selectively blocks IL-36-induced NF-κB activation in NCI/ADR-RES ovarian epithelial cells but does not inhibit IL-1β-induced NF-κB activation or cytokine release in IL-36R knockout NCI/ADR-RES cells or human primary cells[1].
