TLR7 agonist 20 is a selective Toll-like receptor 7 (TLR7) agonist with an EC50 of 0.22 μM. TLR7 agonist 20, as an immunostimulant and vaccine adjuvant, enhances the production of anti-spike protein antibodies, increases IgG2b and IgG2c levels, and thereby drives Th1-type immune responses in mice. In Vivo

TLR7 agonist 20 (Compound 23) (10 μg; i.m.; 2 doses) acts as a potent vaccine adjuvant in mice, inducing a strong Th1 response when administered alone with SARS-CoV-2 spike antigen and enhancing balanced Th1/Th2 responses when co-formulated with alum[1].

Animal Model: C57BL/6 (female, 6-8 weeks old)[1]
Dosage: 10 μg (co-formulated with SARS-CoV-2 spike antigen); 10 μg (co-formulated with SARS-CoV-2 spike antigen and 50 μg alum)
Administration: i.m.; administered at week 0 and week 2
Result: Induced significantly higher anti-spike IgG2b (OD450 ≈ 0.9) and IgG2c (OD450 ≈ 0.8) levels compared to spike antigen alone.
Induced significantly higher anti-spike IgG (OD450 ≈ 2.5), IgG1 (OD450 ≈ 3.0), IgG2b (OD450 ≈ 0.7), and IgG2c (OD450 ≈ 0.8) levels compared to spike antigen alone when co-formulated with alum and spike antigen.
