Toripalimab is a selective, recombinant, humanized monoclonal antibody against PD-1. Toripalimab is able to bind to PD-1 and block the interaction with its ligands. Toripalimab has exhibited primary anti-tumor effects in tumors such as melanoma, lung cancer, digestive tract tumors, hepatobiliary and pancreatic tumors, neuroendocrine neoplasms, nasopharyngeal carcinoma and urothelial carcinoma.

Toripalimab specifically binds to PD-1 (EC50 21 nmol/L) and inhibits binding of PD-1 to PD-L1 [IC50 3.0 nmol/L (≈ 0.5 μg/mL)] and PD-L2 (IC50 3.1 nmol/L) in 293F cells transfected with PD-1 plasmids[2].
Toripalimab (0.01–10 μg/mL) dose-dependently promotes human T-cell proliferation[2].
Toripalimab promotes interferon-γ and TNF-α secretion in activated human peripheral blood mononuclear cell (PBMC)-derived T cells[2].

Animal Model:A genetically modified mouse strain with human PD-1 knocked in (were subcutaneously inoculated with 1 × 106 MC38 cells)[3]

Result:Inhibition of tumor growth was observed in a dose-dependent manner with substantial antitumor efficacy in 1, 3, and 10 mg/kg treatment groups. Decreased significantly at the end of the observation period (day 23)

Immobilized PD-1 Protein, Human (143a.a, HEK293, His, HY-7396) can bind Toripalimab. The EC50 for this effect is 7.504 ng/mL. -
Flow cytometric analysis of 1X106 10 ng/mL PMA and 1 μg/mL Ionomycin co-stimulated Jurkat cells(24h) labeling Toripalimab (HY-P9978, red). Cells were fixed with 4% paraformaldehyde. Then stained with the primary antibody at 1/200 dilution for an hour at 4℃. Alexa Fluor 488-conjugated AffiniPure Goat Anti-Human IgG H&L (HY-P83776) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Human IgG4 kappa (HY-P99003, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black).
