Description

Z56-L23 is a conjugate of RAS-targeting ADC cytotoxic payload-linker with anti-tumor activity. Z56-L23 can be conjugated with HER3 antibody, EGFR antibody or EGFRxHER3 bispecific antibody to form intact antibody-drug conjugates (ADCs). ADC molecules related to Z56-L23 effectively inhibit the proliferation of tumor cells and also significantly suppress tumor growth in xenograft mouse models. Z56-L23 can be used in the research of pancreatic cancer[1].

In Vitro

Z56-L23 conjugated to HER3 antibody, EGFR antibody, or EGFRxHER3 bispecific antibody forms ADCs with high SEC monomer purity (97.70%-98.62%) and DAR values ranging from 4.17 to 7.93[1].
Z56-L23-HER3 ADC and Z56-L23-EGFR ADC potently inhibit the proliferation of NCI-H441 and AsPC-1 cells, with IC50 values ranging from 0.03 nM to 1.60 nM[1].

In Vivo

Z56-L23-HER3 ADC and Z56-L23-EGFR ADC (5-10 mg/kg; intravenous injection; once weekly ×2) exhibit potent anti-tumor activity in NCI-H441 xenograft mice, with tumor growth inhibition (TGI) ranging from 99.15% to 124.66%, and only mild to moderate body weight changes[1].
