Description

ZY39 is a SaClpP agonist. ZY39 promotes the enzymatic hydrolysis of SaClpP and HsClpP in vitro. ZY39 inhibits the growth of Staphylococcus aureus strains. ZY39 reduces the load of Staphylococcus aureus in organs and ascites in mouse peritonitis models and zebrafish infection models, and improves the survival rate of model animals. ZY39 can be used in studies related to Staphylococcus aureus infection, such as peritonitis[1].

In Vitro

ZY39 potently activates SaClpP-mediated α-casein hydrolysis in vitro with an EC50 of 1.1 μM, and exhibits agonistic activity during the hydrolysis of fluorescein isothiocyanate-casein (FITC-casein) by S. aureus ClpP protease (SaClpP), with an EC50 of 0.33 μM[1].
ZY39 activates the hydrolysis of α-casein by HsClpP in vitro with an EC50 of 3.6 μM, but exhibits extremely low activity against gut microbial ClpP (EcClpP, LaClpP)[1].
ZY39 potently inhibits the growth of both drug-sensitive and multidrug-resistant S. aureus strains in vitro. Its activity depends on functional SaClpP, with MIC values ranging from 0.031 to 0.13 μg/mL[1].

In Vivo

ZY39 (0.5-25 mg/kg; i.p.; single dose) demonstrates superior therapeutic efficacy against S. aureus USA300 infection in zebrafish compared to (R)-ZG197 or vancomycin, respectively[1].
ZY39 (25-50 mg/kg; i.p.; single dose) achieves 89-100% survival and reduces bacterial loads by 2-4 log CFU/g tissue in mice with S. aureus USA300-induced peritonitis[1].

Animal Model: Zebrafish (7 to 11 months old, 300 mg, regardless of gender)[1]

Select the appropriate dissolution method based on your experimental animal and administration route.

For the following dissolution methods, please ensure to first prepare a clear stock solution using an In Vitro approach and then sequentially add co-solvents:
To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for in vivo experiments, it is recommended to prepare freshly and use it on the same day.
The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.

  • Protocol 1

    Add each solvent one by one:  10% DMSO    40% PEG300    5% Tween-80    45% Saline

    Solubility: ≥ 2.5 mg/mL (4.56 mM); Clear solution

    This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).

    Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.

    Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
  • Protocol 2

    Add each solvent one by one:  10% DMSO    90% (20% SBE-β-CD in Saline)

    Solubility: 2.5 mg/mL (4.56 mM); Suspended solution; Need ultrasonic

    This protocol yields a suspended solution of 2.5 mg/mL. Suspended solution can be used for oral and intraperitoneal injection.

    Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.

    Preparation of 20% SBE-β-CD in Saline (4°C, storage for one week): 2 g SBE-β-CD powder is dissolved in 10 mL Saline, completely dissolve until clear.
