L-NIO dihydrochloride is a potent, non-selective and NADPH-dependent nitric oxide synthase (NOS) inhibitor, with Kis of 1.7, 3.9, 3.9 μM for neuronal (nNOS), endothelial (eNOS), and inducible (iNOS), respectively[1][2]. L-NIO dihydrochloride induces a consistentfocal ischemic infarctin rats[2].

In Vitro

L-NIO is a potent, non-selective and NADPH-dependent nitric oxide synthase (NOS) inhibitor, with Kis of 1.7, 3.9, 3.9 μM for neuronal (nNOS), endothelial (eNOS), and inducible (iNOS), respectively[1].

In Vivo

L-NIO (2.0 μmol, 3 days post-ischemia) causes focal cerebral ischemia in the adult rat brain[2].

Animal Model: Adult male Sprague Dawley rats (250-350 g)[2]
Dosage: 0.04-2.0 μmol/(3.0-5.0 μL) per rat
Administration: Injected into striatum 3 days post-ischemia
Result: 2.0 μmol L-NIO produced an infarct significantly different in volume from sham animals.
