Nylidrin hydrochloride (Buphenine hydrochloride) is an orally active β-adrenergic agonist. Nylidrin hydrochloride antagonizes NR1A/2B NMDA receptors (IC50 = 0.18 μM in Xenopus oocytes). Nylidrin hydrochloride reduces the levels of NP, HA, and M1. Nylidrin hydrochloride has antiviral activity against multiple H1N1 subtype influenza A viruses. Nylidrin hydrochloride improves hemorrhagic shock and anti-allergic effects.

Biological Activity
IC50: 0.18 μM in Xenopus oocytes (NR1A/2B NMDA receptor)[3]
Nylidrin (4.5-120 μM; 10-35 min) hydrochloride induces dose-dependent and reversible increases in the amplitude of the electroretinogram (ERG) b-wave, decreases in the c-wave, and configurational changes in the optic nerve compound action potential, without affecting perfusate flow or retinal vessel diameter in isolated, arterially perfused cat eyes[2].
Nylidrin hydrochloride potently antagonizes NR1A/2B NMDA receptors (IC50 = 0.18 μM) in Xenopus oocytes[3].
Nylidrin hydrochloride exhibits EC₅₀ values of 7.2 μM against A/Puerto Rico/8/1934 (H1N1) and 12.1 μM against A/Hong Kong/8/1968 (H3N2), with a CC50 of 549.2 μM in MDCK cells[4].

Result:Reduced the levels of NP, HA, and M1 in a dose-dependent manner.
Nylidrin (10 mg/kg) hydrochloride completely prevents mortality (100% survival vs. 0% in virus-only group) and body weight loss in BALB/c mice infected with mouse-adapted influenza A virus (maPR8, H1N1)[4].
Nylidrin (200 mg/kg; p.o.; twice a day) hydrochloride does not improve mortality in maPR8-infected mice but moderates the slope of body weight loss and increases the mean survival time by 1.4 days compared with the virus-only group[4].
