In Vivo

Anti-Human/Mouse/Monkey CXCR3 Antibody (9C5) (2-5 mg/kg; i.v.; single/weekly; 7-21 days) increases pancreatic islet allograft survival in mice, with a single 5 mg/kg dose boosting mean graft survival time from 17 to 33 days, and weekly 2 mg/kg doses achieving 35 days MST with long-term survival in some animals[1].
Anti-Human/Mouse/Monkey CXCR3 Antibody (9C5) (0.5-10 mg/kg; i.v.; single dose) dose-dependently depletes CXCR3-expressing CD4+, CD8+, and B cells in mice, with complete depletion at doses ≥ 2 mg/kg lasting for more than 3 weeks[1].

Animal Model: C57BL/6 (streptozotocin-induced diabetic, pancreatic islet allograft recipients)[1]
Dosage: 5 mg/kg (single pre-transplant dose); 2 mg/kg (weekly post-transplant doses); 2 mg/kg (weekly post-transplant + daily rapamycin for 7 days)
Administration: i.v.; single dose (4-5 hours pre-transplant); i.v.; weekly (days 0,7,14,21 post-transplant); i.v.; weekly (days 0,7,14,21 post-transplant) + p.o.; daily (rapamycin, 7 days post-transplant)
Result: Increased mean survival time (MST) of islet grafts from 17 to 33 days.
Increased MST of islet grafts to 35 days, with 2 out of 7 mice showing graft survival >100 days.
Resulted in 50% graft survival at 60 days.

Reduced effector CD4+ T-cells (CD44high/CD62Llow) and memory CD4+ T-cells (CD44high/CD62Lhigh) from 27.1% to 13.5% relative to isotype control-treated mice after a single 5 mg/kg dose.
