Description
In Vitro
DCG04 reveals a progressive increase in cathepsin activity during RIP1-Tag2 tumorigenesis, identifying cathepsins B, C, L, and Z as active proteases in tumors[1].
DCG04 profiling of flow-sorted RIP1-Tag2 tumor cells shows that infiltrating innate immune cells are a significant source of cathepsin activity, with distinct activity profiles among endothelial, immune, and tumor cell fractions[1].
DCG-04 (50 μM) is an activity-dependent affinity label that covalently modifies multiple active cysteine proteases in DC2.4 dendritic cell extracts at pH 5.5[2].
DCG-04 affinity labeling enriches a 27-kDa cysteine protease from bovine chromaffin granules that is responsible for PE-processing activity[3].
DCG04 activity labeling confirms that immunodepletion and calpastatin loading abolish calpain-1 activity in resealed erythrocytes, which is restored by calpain-1 reconstitution[8].
DCG04 (4 μM; 4 h) is a component of the biotinylated probe cocktail used for ABPP-MS to display active cysteine proteases in N. benthamiana apoplastic fluid, revealing that bacterial infection reduces the activity of nine cysteine proteases[11].
DCG04 (~48 hours postinfection) labels a single ~80 kD membrane-associated protein in P. falciparum-infected erythrocytes, identified as human calpain-1[8].

Recombinant antibody expression and Protein A/G purification
Recombinant antibody expression produces antibody chains from cloned antibody genes in mammalian cells, commonly HEK293-derived cells, so that secreted IgG or Fc-fusion antibodies can be collected from culture supernatant. Protein A/G purification is affinity chromatography in which immobilized Protein A, Protein G, or Protein A/G binds the antibody Fc region; nonbound culture components are washed away, and bound antibody is recovered by changing buffer conditions, commonly acidic elution followed by neutralization.

Protocol For Protein Expression And Purification
Recombinant protein expression in Escherichia coli followed by purification of a His-tagged soluble protein by immobilized metal affinity chromatography (IMAC), with optional MBP fusion and TEV tag removal when the construct includes these elements. The biological readout is production of the encoded target protein, detected as an inducible band at the expected molecular mass by SDS-PAGE and quantified by total protein assay or chromatographic absorbance; the purification readout is enrichment of the target protein in elution fractions after selective binding of polyhistidine residues to immobilized Ni2+/metal-chelate resin and elution by imidazole-containing buffer. Expression is driven by an inducible bacterial expression system, commonly T7/lac-based, in which IPTG or lactose/auto-induction activates transcription and translation of the cloned gene; lower induction temperature, lower inducer concentration, induction timing, and solubility-enhancing fusion tags can influence the frac
