8RK64 is a UCHL1 inhibitor that covalently binds the active-site cysteine residue to form an isothiourea adduct, with selectivity over UCHL3, UCHL5, and other cysteine DUBs. 8RK64 inhibits human UCHL1 with an IC50 of 0.32 μM. 8RK64 is an azide-containing precursor usable as a two-step activity-based probe via click chemistry. 8RK64 can be used for the research of Parkinson's disease.

8RK64 (0.05-200 μM; 30 min) inhibits UCHL1 in an in vitro DUB biochemical activity assay with an IC50 of 0.32 μM; the IC50 values for UCHL3 and UCHL5 are 216 μM and >1 mM, respectively, demonstrating selectivity for UCHL1[1].
8RK64 (5 μM; 24 h) inhibits UCHL1 activity in HEK293T cells, and Cy5-Ub-PA DUB profiling shows reduced residual intracellular UCHL1 deubiquitinase activity[1].
8RK64 (5 μM; 1 h) inhibits UCHL1 activity in HEK293T cell lysates; other DUB bands remain unchanged in Cy5-Ub-PA profiling, demonstrating its selectivity relative to other DUBs[1].
8RK64 (140 μM; 30 min) forms a covalent complex after incubation with recombinant human UCHL1, and LC-MS analysis confirms the covalent binding of 8RK64 to UCHL1[1].
8RK64 (5 μM; 24 h; CuAAC with biotin-alkyne after lysis) makes UCHL1 the most enriched protein relative to the DMSO control in a two-step ABP pull-down/proteomics experiment in HEK293T cells; PARK7 is another enriched protein detected simultaneously[1].

EdU Incorporation Assay (Click Chemistry-Based DNA Synthesis Measurement)
The EdU incorporation assay measures DNA synthesis by adding the thymidine analog 5-ethynyl-2′-deoxyuridine to cells or tissues, where it is incorporated into newly synthesized DNA during S phase. Incorporated EdU is detected by copper-catalyzed azide-alkyne cycloaddition, in which a fluorescent azide covalently reacts with the ethynyl group on EdU, allowing S-phase cells to be detected by fluorescence microscopy, flow cytometry, or high-content imaging. EdU detection does not require DNA denaturation or anti-BrdU antibody access, which preserves sample structure and improves compatibility with immunostaining and multiparameter cytometry compared with BrdU-based detection. EdU can be cytotoxic in a cell-type- and exposure-dependent manner, so pulse duration, concentration, and continuous-labeling designs should be validated for each cell type.
