DCG04 is a biotinylated activity-based probe. DCG04 labels calpain-1 and is applicable for affinity purification and mass spectrometry identification. DCG04 is used in cancer and malaria research^[1]^[2]^[3]^[4]^[5]^[6]^[7]^[8]^[9]^[10]^[11]^[12].

In Vitro

DCG04 reveals a progressive increase in cathepsin activity during RIP1-Tag2 tumorigenesis, identifying cathepsins B, C, L, and Z as active proteases in tumors^[1].
DCG04 profiling of flow-sorted RIP1-Tag2 tumor cells shows that infiltrating innate immune cells are a significant source of cathepsin activity, with distinct activity profiles among endothelial, immune, and tumor cell fractions^[1].
DCG-04 (50 μM) is an activity-dependent affinity label that covalently modifies multiple active cysteine proteases in DC2.4 dendritic cell extracts at pH 5.5^[2].
DCG-04 affinity labeling enriches a 27-kDa cysteine protease from bovine chromaffin granules that is responsible for PE-processing activity^[3].
DCG04 activity labeling confirms that immunodepletion and calpastatin loading abolish calpain-1 activity in resealed erythrocytes, which is restored by calpain-1 reconstitution^[8].
