XY-06-005 (compound 3) potently binds to mutant bromodomains (BRD4 BD1 L94V, BRD4 BD2 L387V, BRD2 BD2 L383V) with IC₅₀ values ranging from 62 nM to 231 nM. In contrast, it shows significantly weaker binding affinity to wild-type BRD4 BD1 (39 nM) and BRD4 BD2 (863 nM), indicating its selectivity for mutant bromodomains over wild-type counterparts^[1].
XY-06-005 can serve as the target protein ligand structure of XY-06-007 (HY-145226). XY-06-007 is a potent and selective B&H-PROTAC BRD4 BD1 L94V degrader with a DC₅₀ of 10 nM^[1].
