Deoxythymidine-5'-triphosphate can be used for DNA amplification in the DNA polymerase chain reaction (PCR). Deoxythymidine-5'-triphosphate is also applicable to research related to diseases such as leukemia.

Deoxythymidine‑5'-triphosphate (dTTP) shows an average concentration of 2.5 pmol/106 cells in resting human lymphocytes; dTTP concentration rises gradually during 24‑72 h culture of PHA‑stimulated human lymphocytes and reaches an average of 9.4 pmol/106 cells at 72 h[1].
Deoxythymidine‑5'-triphosphate exhibits an average concentration of 1.4 pmol/106 cells in normal bone marrow nucleated cells, while its average concentrations in bone marrow nucleated cells corresponding to acute myeloid leukemia, chronic myeloid leukemia, and myeloproliferative disorders are 5.8 pmol/106 cells, 3.8 pmol/106 cells, and 5.9 pmol/106 cells, respectively[1].
Deoxythymidine-5'-triphosphate can be used as a key variable in optimizing PCR amplification strategies for long-fragment, high A+T content mitochondrial DNA: increasing the concentrations of dATP and dTTP in the PCR reaction system while decreasing the concentrations of dGTP and dCTP[2].

EdU Incorporation Assay (Click Chemistry-Based DNA Synthesis Measurement)
The EdU incorporation assay measures DNA synthesis by adding the thymidine analog 5-ethynyl-2′-deoxyuridine to cells or tissues, where it is incorporated into newly synthesized DNA during S phase. Incorporated EdU is detected by copper-catalyzed azide-alkyne cycloaddition, in which a fluorescent azide covalently reacts with the ethynyl group on EdU, allowing S-phase cells to be detected by fluorescence microscopy, flow cytometry, or high-content imaging. EdU detection does not require DNA denaturation or anti-BrdU antibody access, which preserves sample structure and improves compatibility with immunostaining and multiparameter cytometry compared with BrdU-based detection. EdU can be cytotoxic in a cell-type- and exposure-dependent manner, so pulse duration, concentration, and continuous-labeling designs should be validated for each cell type.
