Etevritamab (AMG-596) is a bispecific T-cell engager that targets EGFRvIII and CD3. Etevritamab (5 μg/mL) specifically binds to CD3 and EGFRvIII in CHO cells expressing human or cynomolgus monkey EGFRvIII[1].
Etevritamab (0-1000 ng/mL; 48 h) induces T cell activation, cytokine (IFN-γ) release, and upregulates CD25 expression in a co-culture system containing U-251 MG/EGFRvIII GBM cells and human or cynomolgus monkey peripheral blood mononuclear cells (PBMCs). It also effectively binds to cynomolgus monkey EGFRvIII and CD3, thereby promoting cell lysis[1].
Etevritamab (0-1000 ng/mL; 48 h) induces upregulation of the T cell activation marker CD25 on CD4+ and CD8+ T cells, and mediates redirected lysis of U-251 MG cells with EGFRvIII transferred via microvesicles[1].

Etevritamab (AMG-596) (0.1-0.5 mg/kg; i.v.; daily; 16 consecutive days) significantly prolongs survival in NOD/SCID mice bearing orthotopic EGFRvIII-expressing GBM tumors, with 73% survival at the 0.5 mg/kg dose and 40% survival at the 0.1 mg/kg dose at study end (day 44)[1].

Biological Activity
  • Flow cytometric analysis of 1×106 Jurkat cells labelling CD3E (red) with Etevritamab (HY-P99948). Cells were fixed with 4% paraformaldehyde and permeabilised with 90% methanol. Then stained with the primary antibody at 1/200 dilution for an hour at 4℃. Alexa Goat Anti-Human IgG H&L (AF488) (HY-P83776) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Human IgG1 kappa Isotype Control (HY-P99001, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black).
