GENA-104A16 is a humanized monoclonal antibody targeting CNTN4, with multiple functions including immunostimulation, cytotoxicity and immunoregulation. GENA-104A16 induces CNTN4-positive tumor cell death via multiple mechanisms and enhances T cell-mediated cytotoxicity through immune checkpoint blockade[1].
GENA-104A16 increases T cell-mediated cytotoxicity against CNTN4-expressing human osteosarcoma U2OS cells in co-culture, with this effect dependent on CNTN4 expression by the cancer cells[2].
GENA-104A16 restores T cell proliferation, activation, cytokine release, and cytotoxic killing activity against human CNTN4-expressing cancer cell lines[3].

In Vivo

GENA-104A16 exerts anti-tumor and immune-modulatory effects in CT26/CNTN4 syngeneic mice, increasing total tumor-infiltrated immune cells 2-fold, CD4+ T cells 4-fold, and CD8+ T cells 1.5-fold while reducing Treg cell populations and restoring cytokine secretion[2].
