Description

Gymnoside II is one of the main bioactive constituents isolated from Bletilla striata. Gymnoside II inhibits nano SiO2-induced A549 cell viability reduction, apoptosis, and ROS generation by activating Nrf2. Gymnoside II upregulates HO-1 and γ-GCSc, while downregulating the Bax/Bcl-2 ratio. Gymnoside II is applicable for research on nano SiO2-induced pulmonary injury[1].

In Vitro

Gymnoside II (1–50 µM; 24 h) alleviated the reduction in A549 cell viability induced by nano-level SiO2 and did not affect the proliferation of normal cells [1].
Gymnoside II (25 μM; pre-treatment for 2 h + co-culture for 24 h) inhibits nm SiO2-induced apoptosis in A549 cells by normalizing the expression of apoptotic proteins and reducing the percentage of apoptotic cells [1].
Gymnoside II (25 µM; pretreatment for 2 h followed by co-culture for 24 h) reduces nm SiO2-induced ROS generation in A549 cells[1].
Gymnoside II (25 µM; pretreatment for 2 h followed by coculture for 24 h) upregulates the expression of Nrf2, HO-1, and γ-GCSc and activates the Nrf2 signaling pathway in A549 cells[1].

Cell Viability Assay[1]

Cell Line: A549 cells
Concentration: 1, 10, 25, 50 μM (with or without nm SiO₂)
Incubation Time: 24 h
Result: Elevated the decreased cell viability induced by nm SiO₂ to a relative normal level.
Did not affect normal A549 cell proliferation.

Apoptosis Analysis[1]

Cell Line: A549 cells
Concentration: 25 μM (pretreated for 2 h, then co-cultured with nm SiO₂)
Incubation Time: 24 h (co-culture with nm SiO₂)
Result: Reduced the apoptotic rate from 28.5% to 8.72% and reversed aberrant expression of pro-apoptotic (Bax, Cleaved-caspase 3) and anti-apoptotic (Bcl-2) proteins.
