Description

KDEL is a tetrapeptide serving as an endoplasmic reticulum (ER) retrieval/retention signal. KDEL can target and deliver exogenous antigen peptides to the ER, significantly enhance and prolong MHC class I molecule presentation, and effectively boost anti-tumor immune responses[1].

In Vitro

KDEL (10 μM; 30 min-10 h) fused to the C-terminus of OVA257-264 enhances MHC class I-associated peptide presentation on EL-4 and RMA cells[1].

In Vivo

KDEL (10 μM; subcutaneous injection after DC loading; twice) significantly enhances and prolongs MHC class I presentation of exogenous peptides in C57BL/6 mice, induces a stronger specific CTL response, and significantly inhibits E.G7‑OVA tumor growth[1].

Animal Model: C57BL/6 mice (6-8 weeks old) with E.G7‑OVA tumor challenge[1]
Dosage: 10 μM OVA257‑264‑KDEL peptide
Administration: subcutaneous injection ( days 0 and 7)
Result: Induced greatly enhanced specific CD8+ cytotoxic T lymphocyte (CTL) responses, remarkably prolonged surface MHC class I/peptide complex display, and resulted in significant inhibition of E.G7‑OVA tumor growth in vivo compared with OVA257‑264 or OVA257‑264‑TEWT peptide groups.
