Hexadecyl-CoA

Hexadecyl-CoA is a non-hydrolyzable thioether analog of Palmitoyl-CoA. Hexadecyl-CoA competitively inhibits the acyl-CoA thioesterase activity of HNF-4α with a Ki of 0.3 μM, and inhibits DGAT2 activity with an IC50 of approximately 2 μM. Hexadecyl-CoA also inhibits ATGL activity and serves as a reference inhibitor for DGAT2 LC/MS enzyme activity assays. Hexadecyl-CoA can be used in studies related to lipid metabolism, triglyceride synthesis and lipolysis.

In Vitro
Hexadecyl-CoA (50 μM; 20 min) inhibits TG hydrolase activity of ATGL in COS-7 cell lysates co-expressing ATGL and CGI-58, reducing activity to ~20% of control at 50 μM[1].
Hexadecyl-CoA (1-2.0 μM) potently inhibits the acyl-CoA thioesterase activity of recombinant rat HNF-4α1-LBD (aa132-455) with an apparent Ki of 0.3 μM, via competitive binding that requires its CoA-thioether structure[2].
Hexadecyl-CoA (10 μM) binds specifically to the acyl-CoA site (ACS) of recombinant rat HNF-4α1-LBD (aa132-455) with a stoichiometry of 0.8 nmol per nmol protein, is excluded from the fatty acid pocket (FAP), and requires the intact E-F domain for high-affinity binding[2].
Hexadecyl-CoA (up to 100 μM) does not displace the FAP-bound fluorescent marker C12-Bodipy from human HNF-4α1 recombinant, demonstrating it is excluded from the fatty acid pocket[2].
Hexadecyl-CoA (0.001-100 μM; 60 min) potently inhibits DGAT2 activity in human DGAT2-overexpressing Sf9 insect cell membranes with an IC50 of approximately 2 μM[3].
