ST2-IN-1 is a ST2 inhibitor. ST2-IN-1 blocks the binding interaction between ST2 and IL-33, thereby attenuating the downstream ST2/IL-33 signaling pathway. ST2-IN-1 reduces IL-1β release from mast cells and alleviates ST2 upregulation in cells. ST2-IN-1 can be used for research on inflammatory and immune-related diseases.

Biological Activity
ST2-IN-1 (Compound 31) inhibits the binding of ST2/IL-33 in a cell-free AlphaLISA assay with an IC50 of 7 μM[1].
ST2-IN-1 inhibits ST2/IL-33-mediated NF-κB activation in ST2-overexpressing HEK293 HEKBlue reporter cells, with an IC50 of 7.19 μM[1].
ST2-IN-1 (10-30 μM; 24 h) dose-dependently inhibits IL-1β release from human LAD2 mast cells co-stimulated with IL-33 and 48/80[1].

Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline
Solubility: ≥ 2.5 mg/mL (5.44 mM); Clear solution
This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
