Eupafolin (Nepetin) (6-Methoxyluteolin) is a natural flavonoid isolated from Eupatorium ballotaefolium HBK with potent anti-inflammatory activities. Eupafolin inhibits IL-6, IL-8 and MCP-1 secretion with IC50 values of 4.43 μM, 3.42 μM and 4.17 μM, respectively in ARPE-19 cells.

Pretreatment with Eupafolin dose-dependently inhibited IL-6, IL-8 and MCP-1 secretion with IC50 values of 4.43, 3.42 and 4.17 μM, respectively[1].
Eupafolin (2.5-10 μM; 25 hours; ARPE-19 cells) treatment suppresses IL-1β-induced cytokine (IL-6, IL-8 and MCP-1) expression at mRNA level in ARPE-19 cells[1].
Eupafolin (2.5-10 μM; 1.5 hours; ARPE-19 cells) treatment dose-dependently inhibits phosphorylation of IKKα/β and IκBα, and nuclear translocation of p65. Eupafolin decreases the level of phosphorylated ERK1/2, JNK and p38 MAPK in activated ARPE-19 cells[1].

- To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for In Vivo experiments, it is recommended to prepare freshly and use it on the same day.
- The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.

This protocol yields a clear solution of ≥ 2.08 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (20.8 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.
Preparation of 20% SBE-β-CD in Saline (4°C, storage for one week): 2 g SBE-β-CD powder is dissolved in 10 mL Saline, completely dissolve until clear.
