Nitroxynil is a broad-spectrum veterinary anthelmintic and a substrate of ABCG2. Nitroxynil inhibits ATP production by uncoupling the oxidative phosphorylation process of mitochondria, thereby disrupting the parasite body covering, impairing its motility, and ultimately inducing parasite death. Nitroxynil induces chromosomal aberrations, sister chromatid exchanges and abnormal sperm morphology, and interferes with spermatogenesis. Nitroxynil is widely applicable to studies on fascioliasis, myiasis, Haemonchus spp. infections, and Oestrus ovis infections.

Biological Activity
Nitroxynil (10 μM; 1-4 h) is an efficient in vitro substrate of murine, ovine, bovine, and human ABCG2, as demonstrated by significantly increased basal-to-apical transport ratios (3.25-32.33) in transduced MDCK-II cells compared to parental cells, with transport inhibited by Ko143[1].
Nitroxynil (20 mg/mL; immediate) immediately inhibits muscular contractions in Fasciola gigantica flukes in vitro, causing paralysis and death via oxidative phosphorylation uncoupling[2].
Nitroxynil is metabolized via hydrolysis and nitroreduction in rat liver cytosol in vitro, forming multiple identifiable metabolites including some with flukicidal activity[2].

Reached a 1.8-fold higher plasma area under the concentration-time curve (AUC) in Abcg2−/− mice than wild-type mice (3528.0 µg·h/mL vs. 1922.1 µg·h/mL, p = 0.01).
Exhibited 1.4-fold higher levels in small intestine, 1.7-fold higher accumulation in spleen, and 1.6-fold higher concentration in testis of Abcg2−/− mice compared to wild-type mice at 24 hours post-administration (p = 0.01, p = 0.001, p = 0.02 respectively).
Had concentrations in liver, small intestine, and testis of wild-type mice below the limit of quantification (LOQ) at 96 hours post-administration, while levels in Abcg2−/− mice were 0.7 µg/g (liver), 0.5 µg/g (small intestine), and 1.1 µg/g (testis); showed 3.5 µg/g in kidney of Abcg2−/− mice vs. 0.9 µg/g in wild-type mice (p < 0.001).

This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
