SR8185 (72 h) exhibits potent and broad spectrum antiproliferative activity against a panel of human cancer cells, with IC50s of 1.901 (SCC-9), 0.651 (SCC-15), 1.641 (SCC-25), 0.611 (PC3), 0.532 (DU145),0.550 (LNCaP), 0.463 (Myc-CaP), 0.541 (MCF-7), 1.772 (MDA-MB-231), and 0.613 µM (T-47D), respectively[1].

Cell Viability Determination by MTT Colorimetric Assay
The following protocol uses the MTT colorimetric assay as a classic literature-established method for assessing cell viability/metabolic activity in cultured mammalian cells. MTT[3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide] is reduced by metabolically active cells to a colored formazan product; the amount of formazan is quantified spectrophotometrically and provides an indirect measure of metabolically active viable cells. Importantly, MTT reduction reflects cellular oxidoreductase/metabolic activity rather than an absolute direct count of living cells, so changes in cellular metabolism can alter the signal independently of cell number.
