WX-02-33

WX-02-33 is a TOE1 inhibitor that covalently binds TOE1 and allosterically inhibits its 3′ RNA exonuclease activity. WX-02-33 promotes the binding of TOE1 to spliceosome-associated protein complexes involved in pre-mRNA splicing in a site-specific and stereoselective manner. WX-02-33 exhibits good proteome selectivity in cancer cells, binding only TOE1_C80 and a few other cysteines. WX-02-33 can be used for research on Merkel cell carcinoma.

WX-02-33 (20 μM; 3 h) binds TOE1_C80 with over 80% occupancy in Ramos cells and acts selectively on cysteine across the whole proteome^[1].
WX-02-33 (20 μM; 6 h) inhibits deadenylation of WT-TOE1 but not the C80S-TOE1 mutant in immunoprecipitates derived from MCC142^[1].
WX-02-33 (20 μM; 6 h) promotes the binding of TOE1 to spliceosome-related complexes in MCC142 cells^[1].
WX-02-33 confers a greater advantage on C80S-TOE1 than on WT-TOE1 in the mixed MCC142 competition assay^[1].
