1-Methyladenine is a gonad maturation-promoting regulator. 1-Methyladenine is produced in testes and ovarian follicle cells of starfish under the induction of gonad-stimulating substance (GSS). 1-Methyladenine promotes starfish oocyte maturation and spawning, and modifies bases that regulate DNA structure. 1-Methyladenine converts T-A base pairs in double-stranded DNA into non-disruptive T (anti)m1A (syn) Hoogsteen conformation. If this conformational base is not repaired in a timely manner, 1-Methyladenine transforms into cytotoxic DNA damage and blocks the replication process.

Biological Activity
1-Methyladenine (250 μg dry nerve/mL GSS; 1 h; 22°C) is generated in the non-sperm testicular fraction (Fraction A) of Asterina pectinifera starfish under the action of GSS. After incubation at 22°C with 250 μg dry nerve/mL GSS for 1 h, the average yield per gram of original testis reaches 0.22 μg[1].
When incubated with 250 μg of dry nerve/mL GSS for 2 h, the free cell fraction isolated from testes of Asterina pectinifera produces more 1-Methyladenine (0.26 μg per gram of original testis tissue) than the testis wall fraction, following a prior incubation with 1-Methyladenine (2 h; 22°C)[1].

Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline

This protocol yields a clear solution of ≥ 1.39 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (13.9 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
Add each solvent one by one: 10% DMSO 90% (20% SBE-β-CD in Saline)
Solubility: ≥ 1.39 mg/mL (9.32 mM); Clear solution
This protocol yields a clear solution of ≥ 1.39 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (13.9 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.
Preparation of 20% SBE-β-CD in Saline (4°C, storage for one week): 2 g SBE-β-CD powder is dissolved in 10 mL Saline, completely dissolve until clear.
