Antitrypanosomal agent 33 (compound 1) potently inhibits T. cruzi, T. b. brucei, and T. b. rhodesiense with IC₅₀ values of 0.23, 0.59, and 0.81 μM, respectively^[1].
Antitrypanosomal agent 33 shows low cytotoxicity in MRC-5 cells, with a CC₅₀>61 μM^[1].
Antitrypanosomal agent 33 (100 μM) is a substrate for Trypanosoma cruzi type I nitroreductase, with an observed rate constant of 0.11/s^[1].
Antitrypanosomal agent 33 has favorable human metabolic stability (5.6 mL/min/g liver) and good MDCK membrane permeability (205 nm/sec) but limited aqueous solubility (13 μM)^[1].
