Ara-ATP trisodium
Ara-ATP trisodium is an ATP-competitive poly (A) polymerase (poly (A) polymerase) inhibitor with a Ki value of 4 μM for the chromatin-bound form and a Ki value of 60 μM for the nucleoplasmic (free) form. Ara-ATP trisodium blocks pre-mRNA 3' end processing by inhibiting endonucleolytic cleavage, poly (A) addition, and the formation of cleavage and polyadenylation-specific RNA-protein complexes. Ara-ATP trisodium selectively inhibits initial polyadenylation rather than poly (A) elongation. Ara-ATP trisodium is used in the study of viral infections.

Description
In Vitro
Ara-ATP (0.25-2 mM; 60 min) trisodium inhibits the polyadenylation of SV40L pre-mRNA in HeLa nuclear extracts in a dose-dependent manner, with an inhibition rate of 85% at 0.5 mM and complete inhibition at 2 mM[1].
Ara-ATP (1.0-2.0 mM; 1 h) trisodium completely impairs 3' end processing of adenovirus L3 pre-mRNA in HeLa nuclear extracts at 1 mM[1].
Ara-ATP (0.5-2.0 mM; 30 min) trisodium inhibits endonucleolytic cleavage of SV40L pre-mRNA in HeLa nuclear extracts, with complete inhibition at 2 mM[1].
Ara-ATP (0.5-2.0 mM; 1 h) trisodium inhibits the polyadenylation of "pre-cleaved" SV40L pre-mRNA in HeLa nuclear extract in a dose-dependent manner, with complete inhibition at 2 mM[1].
Ara-ATP (2 mM; 30-90 min) trisodium inhibits the formation of post-cleavage and polyadenylation-specific RNA-protein complexes during SV40L pre-mRNA 3'-end processing in HeLa nuclear extracts[1].
Ara-ATP (1-200 μM) trisodium competitively inhibits rat liver chromatin-associated poly (A) polymerase with a Ki of 4 μM, and inhibits nucleoplasmic poly (A) polymerase with a Ki of 60 μM[2].

RNA extraction experimental
By lysing cells, releasing RNA, and removing impurities such as proteins and DNA, high-purity RNA products are finally obtained. The commonly used traditional method is the guanidine isothiocyanate/phenol/chloroform method (Trizol), which is suitable for a variety of animal materials including animal tissues, microorganisms, cultured cells, etc., and most plant materials.
