    Bone marrow mononuclear cells (BMMCs) from acquired resistant PCM patients were used in Dara-induced ADCC assays. CD138-positive plasma cells were quantified by flow cytometry. BMMCs were treated with Dara (5 or 10 μg/mL) ± inhibitors (Belimumab [BAFF], 4-IPP [MIF], and MYCi975 [MYC]) for 24 h, then co-cultured with NK-cells at an effector:target (E:T) ratio = 1:1 for 12 h. The amount of released LDH was determined using a cytotoxicity assay kit to calculate the % of lysed cells.

    Biological Activity
    • Immobilized BAFF/TNFSF13B Protein, Human (CHO, HY-P7003) can bind Belimumab. The EC50 for this effect is 12 ng/mL.
    • Flow cytometric analysis of 1.5×106 THP-1 cells with Belimumab (HY-P9952, red). Cells were fixed with 4% paraformaldehyde and permeabilised with 90% methanol. Then stained with the primary antibody at 1/200 dilution for an hour at 4℃. Alexa Goat Anti-Human IgG H&L (AF488) (HY-P83776) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Human IgG1 lambda2 Isotype Control (HY-P990096, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black).
