2′-OMe-UMP
2′-OMe-UMP is a primer extension terminator. When 2′-OMe-UMP is present at the -1 position of the primer chain, its 2′-OMe group sterically clashes with the base of the next NTP at the active site (+1 position). This clash weakens the base-stacking interaction between the +1 position and the -1 position, thereby reducing the binding affinity of the next substrate.
