In Vivo

CAN10 (mCAN10, mouse surrogate of CAN10) (10-20 mg/kg; i.p.; twice weekly; 7 weeks) reduces cutaneous and pulmonary fibrosis in a murine cGvHD model of systemic sclerosis, with significant improvements in dermal thickness, hydroxyproline content, myofibroblast counts, and lung Ashcroft scores[1].

Animal Model: B10.D2 (H-2d); BALB/c (H-2d) (allogeneic bone marrow transplantation-induced cGvHD)[1]
Dosage: 20 mg/kg (first dose); 10 mg/kg (subsequent doses)
Administration: i.p.; twice weekly; 7 weeks
Result: Ameliorated clinical signs of cutaneous cGvHD and reduced mean composite scores.
Ameliorated cGvHD-induced weight loss more effectively than the positive control reference drug.
Reduced dermal thickening, accumulation of hydroxyproline, and myofibroblast differentiation compared with vehicle- or isotype control-treated mice.
Reduced Ashcroft scores and hydroxyproline content in lung tissue compared with vehicle- or isotype control-treated mice, with numerical decreases in collagen-covered lung area.

Biological Activity
  • Flow cytometric analysis of 1X106 HepG2 cells labeling IL1R3 with CAN10 (HY-P991267, red). Cells were fixed with 4% paraformaldehyde and permeabilised with 90% methanol. Then stained with the primary antibody at 1/200 for an hour at 4℃. AF488-conjugated Goat Anti-Human IgG H&L (AF488) (HY-P83776) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Human IgG1 kappa Isotype Control (HY-P99001, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black).
